Assignment Task:
Task 2 is worth 4.5% of the topic(each question will be marked out of 10). Please read the instructions about the take-home exam before submitting your answers to these questions into the task 2 assignment box. The answers to task 2 are due Monday the 30 th March at 10 am. Please check your answers in Turnitin before the final submission. Each of your answers should be no more than 100 words (include diagrams where appropriate/requested, but do not “cut and paste” diagrams from articles provided). You can draw an image by hand and take a photo to insert into your document if you wish. Below is part of a hypothetical coding sequence of a eukaryotic gene showing it contains a phosphate-binding loop or “walker A motif (G-X(4)-GK-{TS}). The Walker-A motif is part of a universal binding pocket found in a vast array of proteins that span all kingdoms of life.
Refer to the attached review article.
i. What is the function of the P-loop motif in the proteins that contain this sequence of amino acids?
ii. What is the most conserved and important residue in this motif and what is its likely specific function?
iii. If your role were to generate an inactive mutant of this protein encoded by this gene (based on the same mutation tested in adenylate kinase reported in the review article), show the primers you would design to generate that mutant.
iv. List the important features of the primer that you have checked to ensure the success of your experiment.
v. Once you have designed your primers, draw out a flow diagram, with a few explanatory notes, showing experimental steps needed to perform this mutagenesis experiment (refer to the lecture notes for help).
